Endotoxin testing: the test a purity number can’t replace

What bacterial endotoxins are, how LAL and recombinant tests measure them in EU, why injectables need them, and why most peptide COAs don't include them.

The short answer ·

Endotoxins are fragments of the outer membrane of Gram-negative bacteria (lipopolysaccharide). Injected, they can cause fever and inflammatory reactions even when no live bacteria are present, and they survive normal sterilization. Labs measure them with the Limulus amebocyte lysate (LAL) test or recombinant factor C assays and report endotoxin units (EU).

A purity or identity test does not detect endotoxin.

What are endotoxins?

Bacterial endotoxins are lipopolysaccharides (LPS) from the outer membrane of Gram-negative bacteria. They are released when bacteria die or divide. They are pyrogens, meaning they cause fever, and in sufficient amounts in the bloodstream they can trigger serious inflammatory reactions. They are heat-stable and are not removed by ordinary sterilizing filtration.

How are endotoxins measured?

FDA’s guidance Pyrogen and Endotoxins Testing: Questions and Answers covers the accepted methods for regulated products:

Method Principle Readout
LAL gel-clot Horseshoe crab blood lysate clots in presence of endotoxin Pass/fail at a sensitivity
LAL turbidimetric Clotting increases turbidity over time Quantitative (EU)
LAL chromogenic Enzyme cascade releases a colored product Quantitative (EU)
Recombinant factor C (rFC) Recombinant enzyme, fluorescent readout, no animal lysate Quantitative (EU)

Results are reported in endotoxin units (EU) per mg or per mL. Peptide samples can interfere with the assay, so labs run spike-recovery controls. Ask whether the report shows them.

Why is endotoxin missing from most COAs?

Most peptide certificates you will see online are purity and sometimes identity reports. Endotoxin is a separate, extra-cost test. MZ Biolabs is explicit that its purity testing “does not test for activity, sterility/endotoxins, heavy metals, or pH.” Freedom Diagnostics lists endotoxin screening as a separate menu item.

Why do regulators care about endotoxin?

FDA’s 2026 warning letters to online peptide sellers include language on this exact risk. Injectable products “are delivered directly into the body, sometimes directly into the bloodstream, and therefore, bypass some of the body’s key defenses against toxins and microorganisms” (March 2026 letters).

How do you read an endotoxin result?

  • Units and basis: EU/mg or EU/mL, and how the sample was reconstituted.
  • Method: LAL kinetic, chromogenic or rFC; gel-clot gives only a threshold.
  • Controls: positive product control (spike recovery) within range.
  • Sample identity: same lot as the purity test?

There is no recognized acceptance limit for research-use products. Limits for approved drugs depend on dose and route. We report methods. We do not set thresholds for human use.

Frequently asked questions

Does sterile mean endotoxin-free?

No. Sterility means no viable microorganisms. Endotoxins are heat-stable fragments of dead bacteria and can remain after sterilization or filtration.

What unit is endotoxin reported in?

Endotoxin units (EU), usually per milligram or per milliliter. Limits for approved drugs depend on dose and route; there is no recognized limit for research-use products.

Why did FDA single out injectables in 2026 warning letters?

FDA wrote that injectable products 'bypass some of the body's key defenses against toxins and microorganisms that can lead to serious and life-threatening conditions.'
